So basically, I’m not affiliated with anyone I know in real life on Tumblr. I used to have two people I know from real life following me, but I blocked them and then changed my URL.
So, would it be fine for me to identify myself as a homosexual publicly on Tumblr? I’m in the closet in real life, and no one knows about my sexuality. However, since no one in real life will be able to see it, should I just identify myself as a homosexual on Tumblr?

In fact most people just assume you are gay in tumblr. “Coming out” on tumblr only applies to straight people who have to come out as straight.
The best part about Tumblr is that you can meet awesome people just like you and blog about whatever your like under an anonymous identity. So… yes reveal your sexual orientation however you like. The Tumblr community welcomes you!
Blocking them doesn’t guarantee that they won’t still see your posts. Your blog will still show up in their following lists, and while they won’t recognise your URL, they can still click on it and look at your blog, and if you haven’t given your blog a thorough makeover, they’ll recognise your blog.
Coming out on Tumblr won’t be an issue, because Tumblr is in general very accepting, of sexuality in particular. But as long as real life friends could access your blog, just know you could accidentally out yourself in real life in the process, and you don’t sound like you’re quite prepared for that.
If you want to be completely sure, make a new blog, and don’t give out any details that could identify you to the people you know in real life, should they happen to stumble across it. (Meaning, don’t post pictures, don’t say where you live and don’t reveal your name.)
