2. Now you want to find the AFP gene in this cod genomic DNA library. Although the genome of the Arctic cod has not been sequenced, from careful study of GenBank, the online database of
DNA sequences, you are able to create a probe for the AFP gene from other arctic fish species. Explain what a probe is, and describe how you will use this to isolate the AFP gene from the
DNA library. Be specific!
3. You have now isolated the bacterial colony which contains the plasmid with a DNA fragment that includes the cod AFP gene. As a research control, you need to double check the size of
that cod DNA fragment. Describe a procedure for cutting out the cod DNA fragment from the plasmid and measuring its size.
4. Once you isolate the bacterial colony containing the plasmid with the cod AFP gene, you need to combine this gene with the jellyfish GFP gene on a single plasmid. As is standard practice in
the biotech industry, the GFP plasmid has a multiple cloning site and a selectable marker. Describe this cloning process. Be specific!
5. Research is going well! You are now working on overcoming the obstacle of getting your recombinant plasmid into the pickling cucumbers and getting high expression of both AFP and
GFP. Unfortunately, one night someone breaks into your lab and steals your most promising prototype cucumber seedlings. Your company’s security force calls in the CSI New York team

I am writing a short story about an orphan. His parents were murdered on the day the sun and the moon lied immobilized in which everything was frozen (earth sun and moon). He goes to an orphanage and fin his best friend, Well afterwords, the main characters brother gets murdered (thirteen years later) and the murderer is his best friend. I like how I outlined things but I have one gigantic writers block. I want the best friend to be the murderer of the family and I would like it to be affiliated with the immobilized moon and sun. I thought of having the dad as the president of an organization that deals with the moon and the sun but I just don’t know how that would fit together. I would really appreciate it if you had any ideas because this has been bothering me for quite some time. I’d like something good and would make someone think oh my god! I’m thinking about applying to doctorate of physical therapy programs. What do you think the growth will be for this profession? I know that BLS and other job related sources say that need for this job will grow very fast, but I just would like to have thoughts from people in medicare.

In 2012 I made $18,000 in self employment income. I am young and have only had 1 real job outside of my home. I was laid off from my last job and needed income fast so I got into affiliate marketing and started making money fast. Soon, I was making more than I thought I would. Recently, I found out that I am suppose to pay taxes on self employment income but I don’t know where to start.
Please help! How much will I owe in self employment tax, federal, state, and penalties all together?
Will I get in big trouble?
How do I go about paying my taxes. I am a little confused so I need as many details as possible.
